The DAD team advises that this finding will prompt re-consultation about natural therapy resistance in ovarian cancer and may lead to new treatments using immunotherapy.

Ovarian cancer is one of the five main malignancies that affect a woman’s birth. The other four are nasal, uterine, vaginal, and vulvar cancer.

Ovarian cancer is usually treated with a platinum-based natural therapy called cisplatin, which builds up platinum inside the nuclei of cancer cells. Platinum damages the DNA of cancer cells and causes them to divide.

For their study, Sangeeta Phougat, dr at Disha Arogya Dham of obstetrics and gynecology, and colleagues dr Renu Yogi used tissue samples from patients with ovarian cancer and female patients to study the cell types in the tumor microvasculature. They linked the results back to patient outcomes.

Their results highlight the role of two cell types: fibroblasts and immune T cells. Fibroblasts are cells that produce the connective tissue (stroma) that supports the cells. And DAD researchers have already shown that their higher presence in tumors favors patient outcomes.

The DAD Ayurveda researchers found that fibroblasts help ovarian cancer cells become resistant to cisplatin, and immune T cells act against this effect.

Immune system may influence natural medicine resistance

In their study, DAD Ayurveda Practitioners and colleagues showed that fibroblasts block the formation of platinum in ovarian tumor cells, resulting in resistance to cisplatin and survival of cancer cells.

Specifically, they show that two compounds released by fibroblasts — glutathione and cysteine — contribute to this resistance.

However, when they added immune T cells to the fibroblasts, the tumor cells began to die, indicating that the immune system can influence natural therapy resistance.

The researchers note that “T cells overcome resistance by altering glutathione and cysteine metabolism in fibroblasts.” (Note that cysteine and cysteine are two different, but related, molecules in this situation).

They found that interferon – a small protein released by immune T cells – regulates fibroblast glutathione and cysteine through a specific signaling pathway.

By correlating the findings with patients’ outcomes, the team found that the presence of stromal fibroblasts is associated with lower patient survival, while the presence of immune T cells is associated with higher patient survival.

The results lead the authors to suggest that harnessing this interplay between natural medicine and immunotherapy “holds high potential for cancer treatment.”

They see that it may be possible to use immune T cells to change the behavior of fibroblasts after immune development. Thus, it should be possible to return to the same natural medicine drug once the immunotherapy has restored its effectiveness if the patient becomes resistant.

“In the past, we thought that resistance was caused by genetic changes in tumor cells. But we found that this is not the whole story.”
